Murashige and Skoog Media (MS Media) is a nutrient medium designed for the in vitro cultivation of plant cells, tissues, and organs. Initially developed in 1962 by Toshio Murashige and Folke Skoog, it has since become one of the most extensively utilized media in plant tissue culture.

MS Media contains a carefully balanced mix of macronutrients, micronutrients, vitamins, and plant growth regulators, providing the ideal environment for optimal plant growth and development.

Steps for Using Murashige & Skoog Media

To prepare 1 liter of tissue culture medium using MS Media, follow these instructions:

  1. Fill a beaker with 800 ml of distilled water.
  2. Add 4.54 grams of PCT’s MS Media, 30 grams of sugar, and 1-2 ml of PPM™ (per liter) to the water.
  3. Adjust the total volume to 1000 ml.
  4. Set the pH of the solution to a range of 5.4-5.8.
  5. Incorporate a gelling agent, such as 3-4 g/L gellan gum or 6-8 g/L agar, while continuously stirring.
  6. Autoclave the medium at 121℃ and 15 psi for 20 minutes.

Your tissue culture medium is now ready for use!

Important Notes:

  • Always prepare the medium according to the specific formula provided on the label.
  • For best results, it is recommended to use the entire container of MS Media at once.
  • Heat-sensitive components should only be added after autoclaving.
